Fabrication of Collagen–Chitosan nanofibrous scaffold with honey underlayer for skin tissue engineering |

Abstract: |
Skin regeneration is an important field of tissue engineering . in larger burns , present treatments are insufficient in preventing scar formation. For this reason , we have fabricated electrospun nanoscaffolds from mixture of chitosan (CHI) (75-85% deacetylated, low molecular weight) and collagen type I (COL) extraction from rat tail tendon . the electrospining process was performed at different ratio of COL and CHI (100:0, 80:20,50:50, 40:60 ) and was selected the best ratio with use of spivak mathematical models. Collagen is a poly protein and has good properties such as having a biological origin, non-immunogenicity, excellent biocompatibility and biodegradability and it provides a favourable surface for cellular attachment. Chitosan can modify the properties of collagen and improve mechanical properties of collagen. In this study, we were produced underlayer of honey with spin coating equipment.use of honey underlayer showed powerful antibacterial efficacy, farthermore it improved and facilitated electrospining process. The results of scanning electron microscopy (SEM) showed morphology of electrospun scaffolds and the best ratio of Chi/Col to obtain a uniform and regular nanofibres. The results of analysis demonstrated that these nanofibre have high swelling ratio , because of high porosity. The consequence of MTS assay showed the increasing of cells and good cell behaviours. Results of this study showed that nanoscaffols of Chi/Col with honey underlayer would be a potential condidate for burn treatment. |
